proteasomesThe American scientists managed to find out the substances oppressing the basic protective mechanism of the originator of a tuberculosis, thus not toxic for human cells. These bonds can become new medicines for a widespread serious infection, informs ScienceDaily.

Earlier the same group of scientists of four research centres in the USA has come to conclusion, that possibility of a micobacterium of a tuberculosis is long to remain in a human body in "a dozing condition" is caused by presence at it proteasomes- the fermental complex "clearing" a bacteriemic cell from fibers, damaged by immune system of the person. Without proteasomes accumulation of such fibers would lead to death of a microbe.

Having found out it, researchers have started search of the substances blocking this protective complex, and, hence, depriving a protection micobacterium. The problem became complicated that in human cells there are similar complexes, and the potential medicine should not oppress their function.

Scientists have spent screening of 20 thousand bonds about their ability to block mycobacterial proteasomes. During screening they have found out group of the potential inhibitors, which activity then have tested on tuberculosis originators, and also on cultures of cells of an epithelium of monkeys and human immune cells.
Two substances from group oksatiazol-2-onov have appeared in one thousand times more active in the proteasomes micobacteria relation, in comparison with proteasomes the person. In experiment they effectively suppressed growth of bacteria without visible toxicity for cells of mammals.

By the structural analysis executed on equipment National Synchrotron Light Source in Brukhevensky laboratory of the Department of Energy of the USA, researchers have established, that these bonds suppress proteasomes activity by change of their active centre and the general molecular conformstion.
The found out bonds are the most perspective potential medicines for a tuberculosis, however the long researches confirming their efficiency and safety are necessary for introduction in clinical practice.
